Microdialysis – Muscle Microdialysis

Microdialysis can be used to measure extracellular levels of amino acids, which can serve as putative biomarkers of protein breakdown and synthesis. Optimized microdialysis protocols also have the potential to be used in clinical settings for evaluation of drugs affecting proteolytic pathways.

chart

Change the y-axis value to AUC (30-240 min) (µmol/L) [not mol/l]. The bar colors should be unified for the pairs (day 1 and day 5) of amino acids.

Amino acid levels in the mouse gastrocnemius muscle. Cannulae were implanted in the left gastocnemius muscle of mice on day 1 and the extracellular levels of several branched amino acids were measured and expressed as the AUC for the 5 hr sampling period. Cannulae were implanted in the right gastrocnemius muscle of the same mice on day 5 and amino acid levels were measured once again. Significant differences were observed between the levels of different amino acids but the level of each amino acid measured on day 1 and 5 remained very stable. N = 4

chart 2

Topical application of 1% capsaicin (horizontal bar) to the abdominal skin of anesthetized rats produced a marked increase in PGE2 levels but almost no effect on histamine levels.
