Microdialysis – Pharmacokinetic/Pharmacodynamic Determinations

Dual and triple probe microdialysis can be used to simultaneously measure brain levels of transmitter and drug along with blood levels of drug and precisely determine the relationship between pharmacokinetics and pharmacodynamics in the brain.

Top - Methylphenidate was injected at time = 0 and the levels of dopamine (DA) and methylphenidate itself were measured simultaneously using separate probes. The blood level of methylphenidate was measured using a third probe. The increase in DA level roughly parallels the increased brain and blood levels of methylphenidate for the first 2 hrs but remains elevated even after the levels of methylphenidate begin to fall Bottom – Cocaine was injected at time = 0 and again at time = 100. The rapid rise and fall in the level of DA very closely parallels both the brain and blood levels of cocaine.

Similar to methylphenidate (Ritalin), d-amphetamine was highly efficacious.
