Animal Model – Autism Spectrum/Developmental Disorders

Autism

The primary diagnostic indicators of autism are abnormal behaviors, rather than biochemical, neuroanatomical or other physiological indices. Determining whether a proposed mouse model for autism recapitulates one or more of the core clinical symptoms can provide valuable insight as to the functional impact of altered genes or environment. Some models, such as the Shank 3 mutant, have construct validity based on studies showing a link between autism and autism spectrum disorder and a genetic mutation.

Ultrasonic Vocalization in Rat Pups

The ultrasonic cries are sensitive to drugs with anxiolytic properties like 5-HT1A receptor agonists (buspirone) and GABAA-receptor agonists (chlordiazepoxide).

chart 2

The Ultrasonic vocalizations were recorded under two temperature conditions (37 degrees C and 18 degrees C), reflecting a low and high stress state.

This paradigm also distinguishes other drug classes, including SSRIs (e.g. fluvoxamine).

Simultaneously other measurements of side effects (negative geotaxis and body temperature) can also be obtained.
